  • Application notes
ELISA: Use at a concentration of 2 µg/ml.
    IHC-P: 1/20 (human) - 1/400 (rat).
WB: Use at an assay dependent dilution. Predicted molecular weight: 43 kDa.

    Not tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
  • Target

    • Function
Contributes to the lung's defense against inhaled microorganisms. May participate in the extracellular reorganization or turnover of pulmonary surfactant. Binds strongly maltose residues and to a lesser extent other alpha-glucosyl moieties.
    • Sequence similarities
      Belongs to the SFTPD family.
      Contains 1 C-type lectin domain.
      Contains 1 collagen-like domain.
    • Post-translational
      modifications
      The N-terminus is blocked.
Hydroxylation on proline residues within the sequence motif, GXPG, is most likely to be 4-hydroxy as this fits the requirement for 4-hydroxylation in vertebrates.
    • Cellular localization
Secreted > extracellular space > extracellular matrix. Secreted > extracellular space > surface film.

    Answer

Thank you for this additional information. A few suggestions just to follow a protocol a little more similar to how we have tested. We generally recommend blocking in 5% milk in TBST and incubating the primary antibody overnight at 4 degrees. You may also wish to try changing your lysis buffer to RIPA if possible.
