Target

  • Function

    Component of the chromosomal passenger complex (CPC), a complex that acts as a key regulator of mitosis. The CPC complex has essential functions at the centromere in ensuring correct chromosome alignment and segregation and is required for chromatin-induced microtubule stabilization and spindle assembly. The complex with RAN plays a role in mitotic spindle formation by serving as a physical scaffold to help deliver the RAN effector molecule TPX2 to microtubules. May play a role in neoplasia. May counteract a default induction of apoptosis in G2/M phase. Inhibitor of caspase-3 and caspase-7. Isoform 2 and isoform 3 do not appear to play vital roles in mitosis. Isoform 3 shows a marked reduction in its anti-apoptotic effects when compared with the displayed wild-type isoform.

    Expressed only in fetal kidney and liver, and to lesser extent, lung and brain. Abundantly expressed in adenocarcinoma (lung, pancreas, colon, breast, and prostate) and in high-grade lymphomas. Also expressed in various renal cell carcinoma cell lines.

    Contains 1 BIR repeat.

    Expression is cell cycle-dependent and peaks at mitosis.

  • Overlay histogram showing HeLa cells stained with ab93274 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab93274, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

I apologize for delay as I was gathering temperature sensitivity data for this antibody. Unfortunately laboratory is not aware of any kind of degradation of this antibody after storing it at -20C. We however acknowledge the time you have spent and would be happy to provide another free of charge replacement. Could you confirm if you are interested?

I have one suggestion which you can follow if you like "store 1-2 aliquots at -20C and to the rest add 1:1 ratio of Glycerol before storing - this will help to understand if freezing at -20C have damaging effect on antibody.

Thank you for your email. I am sorry to hear that you have been experiencing problems with this antibody.

The ab7170 antibody is out of stock which is why it was unpublished. I am sorry we will not be able to provide this product. The product ab93274 is indeed a purified antibody which however will not be affecting the specificity of this antibody.

I have few following further questions about the protocol;

- The image is totally clean so could I double check? have you used 400 cells/ well or 50- 90ug protein/ lane?
- Did you observe same clean membrane after trying 3 times?
- Is the secondary antibody fine? Have you tried "loading control" with the same lysates?
- Is it true no protease inhibitors were added? Could you try after adding a PI cocktail?

Finally would you like to try a different lot of this product.
