    SW480 whole cell lysate
  • General notes

    Cell line: SW480 (Human colorectal adenocarcinoma).
    Growth media: Leibovitz’s L-15 medium with 2 mM L-glutamine, and 10% FBS.


    SW480 cell lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylene diamine tetra acetic acid, 1 mM phenyl methyl sulfonyl flouride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecyl sulfate, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The cell lysate was boiled for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecyl sulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol.

    Thank you for contacting us. We are happy to help customers find the most suitable positive controls for their research purposes; however we are not specialized in this particular field and do not want to recommend a control that may be unsuitable. We encourage customers to consult the latest literature available through PubMed and other resources in order to find the most up-to-date information about their specific research interests. A preliminary literature search showed that human colon adenocarcinoma should be a suitable positive control. A Human colon adenocarcinoma cell line SW480 was also found to be positive for HOXA9. We do offer a SW480 lysate for WB that could then be used as positive control: ab3957 (SW480 (Human colon adenocarcinoma cell line) Whole Cell Lysate) Click here (or use the following: https://www.abcam.com/SW480-whole-cell-lysate-ab3957.html). I also found that the following cells were found positive: 721_B cells and hep2G cells. I am sorry that I could not be more helpful, but I hope that the available literature in this area can provide some clarification.
