    • Function

      Neuronal phosphoprotein that coats synaptic vesicles, binds to the cytoskeleton, and is believed to function in the regulation of neurotransmitter release. The complex formed with NOS1 and CAPON proteins is necessary for specific nitric-oxid functions at a presynaptic level.
    • Involvement in disease

      Defects in SYN1 are a cause of epilepsy X-linked with variable learning disabilities and behavior disorders [MIM:300491]. XELBD is characterized by variable combinations of epilepsy, learning difficulties, macrocephaly, and aggressive behavior.

      Belongs to the synapsin family.
    • Post-translational
      modifications

      Substrate of at least four different protein kinases. It is probable that phosphorylation plays a role in the regulation of synapsin-1 in the nerve terminal. Phosphorylated upon DNA damage, probably by ATM or ATR.

      Thank you for your patience. The IF validation for this antibody was done by a collaborator, so unfortunately we do not have a specific protocol to provide. The neurons were cultured for 14 days. I hope this helps, please let me know if you need any additional information or assistance.

      I've attached the specific IP protocol for ab8, which was optimized with Sacc cells. I am hopeful that this protocol will be a good referece, but if you would like to send some details about your experiment I will also see if there are any other suggestions to make.

      Recently, I've been doing some experiments with some colleagues on synapsin 1.
      I have purchased 2 antibodies from abcam: one N-ter (ab18814) for WB and one (ab8) for IF.

      The ab18814 is working great! Do you think you could tell me the amino acids recognized by this antibody? my colleagues have performed MALDI technique and they need to make sure that the peptide captured by proteomic is the same than the one seen in WB.

      For IF, unfortunately, the ab8 gave poor results on mouse fixed brain sections, with small numbers of neurons stained (ab18814 didn't work on IF neither).
      Do you have any recommendations for another ab to test? Maybe ab64581?

      Regarding ab8, I'm very sorry to hear that it isn't working for you! This should be working in IHC on mouse tissues, so I'm not sure why it is giving you trouble. I took a look through our catalog, and it looks like ab64581 would be a good replacement. Ab64581 has been tested in IHC and reacts with mouse, and has very positive reviews. I've included a link to the datasheet below so you can take a look and make sure this is the one you want. I'm happy to send you a vial free of charge.

      For ab18814, this product is produced by a third party so we do not have the exact immunogen sequence. We do know that the immunogen peptide is taken from around amino acid 12. In my experience, immunogen peptides are normallysomewhere around 15-20 amino acids long, but obviously this could be different for this antibody. I'm going to see how much information I can get from the originator of the antibody, but I am not sure whether they will release the exact sequence to us.

      Thanks for your reply. One example of a popular target for synapse staining is PSD95. A good antibody to detect this protein in a variety of applications and species is ab18258.       I haved got an antibody(rabbit polyclonal to synapsin I--ab8) from you company.The application notes in datasheet tells me that the recommended dilutions for WB is 1:200;but it is used at a concentration of 0.1ug/ml(1:2000)for IHC-F.Normally,we use the higher dilution for WB than for IHC(maybe 1:3000 or higher). So i am not sure if the recommended dilution 1:200 for WB in the datasheet is correct.Please tell me the recommended dilution for WB. Could you please confirm the recommended dilutions for WB?

      I recently ordered a vial of pAb to Synapsin1(Novus), NB300-104 (lot 1650 304kn), 10 ug reconstitute in 50 ul. We tried on frozen sections as well as paraffin sections. Nothing worked! We even use pretty high concentration, 1/20 dilution. Will you kindly help me find a reliable technique to do the work. References will also be most welcome!

      I'm sorry to hear that you are having trouble getting this antibody to work in your research. The antibody is suitable for use in IHC on both frozen and fixed sections and Novus recommend a dilution of 1:1000. There is a reference on the datasheet that features the use of this antibody, but I'm not sure whether or not that was in IHC: Danglot L et al. Association of gephyrin with synaptic and extrasynaptic GABAA receptors varies during development in cultured hippocampal neurons. Mol Cell Neurosci 23:264-78 (2003). PubMed: 12812758. In order to help you further I will need more details of your protocol and the nature of the material on which you are working. The technical help section of our website will be useful to you as it contains many helpful suggestions as to how you could optimise your protocol.
