Specificity
Detects a ~18kD protein in WB, corresponding to predicted size of VAMP from human, mouse, rat, bovine, guinea pig, hamster, monkey, porcine, rabbit, sheep and Xenopus tissue. This antibody detects VAMP1 (amino acids 1-118) and VAMP2 (amino acids 33-96)-GST fusion proteins expressed in E. coli. No reactivity was seen with a ~18kD protein in mouse liver extracts. The clone number has been updated from (4E240) to (SP10 ) both clone numbers name the same antibody clone. -

Relevance
Synaptobrevins are highly conserved between vertebrates and invertebrates. It is a major constituent of synaptic vesicles and peptidergic secretory granules in all neurons examined so far. In addition, it is present on secretory granules of neuroendocrine cells. Synaptobrevins, Syntaxins, and the 25 kDa synaptosomal associated protein SNAP25 are the main components of a protein complex involved in the docking and/or fusion of synaptic vesicles with the presynaptic membrane. -

Cytoplasmic vesicle › secretory vesicle › synaptic vesicle membrane. Single-pass type IV membrane protein. Cell junction > synapse > synaptosome. Isoform 3 is found in mitochondria, mitochondrial outer membrane. -
