  • ab14692 staining Synaptophyson in mouse retina (8 weeks old) and brain tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). 10µm frozen sections were fixed with 4% paraformaldehyde for 10 minutes and blocked with 10% donkey serum for 20 minutes at room temperature. Samples were incubated with primary antibody (1/100 in PBS) overnight at 4°C. An Alexa Fluor 488®-conjugated donkey anti-rabbit IgG polyclonal (1/1000 in PBS) was used as the secondary antibody (1 hour at room temperature). Counterstained with Hoechst 33258 (blue) for 10 minutes at room temperature.

Images shows: GCL - ganglion cell layer, IPL - inner plexiform layer, INL - inner nuclear layer, OPL - outer plexiform layer, ONL - outer nuclear layer; DG - dentate gyrus region of the hippocampus.

  • Synaptophysin antibody (ab14692) western blot on mouse forebrain extract (10µg protein/lane). The blot was exposed for 10 minutes to autoradiographic film. Ladder markers from 7-194 kDa.

    Lane 1 = 1/500 ab14692
    Lane 2 = 1/200 ab14692

  • ab14692 staining cultured ED18 rat primary hippocampal neurons by ICC/IF. The cultured cells were fixed with 4% paraformaldehyde for 5 minutes and blocked with 10% donkey serum in 0.1% PBS-0.3% TritonX for 30 minutes at 24°C. The cultured cells were then stained with ab14692 at 1/200 in 0.3% TritonX with 0.1% PBS and 10% donkey serum for overnight at 4°C. An Alexa Fluro 488 donkey anti-rabbit polyclonal antibody at 1/1000 was used as the secondary antibody. Nuclei were stained with 1.43µM Hoechst and can be observed in blue. Synaptophysin expressed in axon, dendrites and particularly carrying in the vesicles.

Answer

Thank you for that confirmation.

I have now arranged for a replacement vial to be sent to you. This is on the order number xxxxx(Purchase order number xxxxxx). This should be with you on Wednesday. If you have any problems receiving it please do let me know.

If you continue to have problems with the antibody please also let me know. As discussed in my previous email, there are a few protocol changes that I think would be worthwhile trying in order to see if the antibody performs any better with your samples.

I look forward to hearing how you get on. Until then, I wish you all the best with your research.

Question
Answer

Thank you for providing that information.

From having reviewed the protocol details provided there are q few thinks I would suggest would be worthwhile to try to obtain the best staining with the antibody.

1. I would suggest increasing the concentration of antibody used, trying 1/100 to start with to see if any improvement is seen.

2. It may be worthwhile to try and optimise the antigen retrieval used. Applying sodium citrate buffer for 30 minutes bay be a little too long. I would suggest treating the tissue with boiling citrate buffer (using microwave or pressure cooker) for 5, 10 and 20 minutes to see which gives the best results.

I do however understand that you have spent some time in optimising the conditions and you have used the antibody previously. I would therefore be happy to provide you with a replacement vial of ab14692 (of a different lot number), or an alternative antibody altogether such as rabbit monoclonal https://www.abcam.com/Synaptophysin-antibody-EP1098Y-ab52636.html.

If you would like for me to arrangeeither of these options, could you pleaseconfirm the delivery address andbilling address (as we only have that of ATOM which placed the order originally).

I thank you for your cooperation and look forward to receiving your reply.

Answer

Thank you for providing that information. That has clarified the situation encountered by the customer very well. Could I ask for a few more details to be provided:

1. Which type of tissue samples are being stained? Human? Mouse? From which organ?

2. Has the secondary antibody been used successfully before?

3. Do you have the order number on which the ab14692 was purchased?

I look forward to receiving your reply.

Question
Answer

Thank you for contacting us.

Here please find the information about our testing discount program. Below I also include information about our Abpromise guarantee.

For UNTESTED species and/or applications, we have established a testing discount program. Here is a brief description of how it works:
The testing discount program is for customers who like to use an antibody/protein on an untested species/application. You would purchase the antibody at full price, test it and submit an Abreview with your data (positive or negative). On your next order you will receive a discount for ONE antibody at the full price (100%) of the antibody you have tested. The terms and conditions applicable to this offer can be found here: https://www.abcam.com/collaborationdiscount.

Abpromise guarantee:

We guarantee all our products for TESTED species and applications as listed on the datasheet. The guarantee is valid for 6 months after a purchase. For more details about our Abpromise guarantee please check this link: https://www.abcam.com/abpromise.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
