  • ab8049 staining Synaptophysin in mouse hippocampus tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with formaldehyde and blocked with 5% BSA for 1 hour at 24°C. Samples were incubated with primary antibody (1/100 in BSA) for 12 hours at 4°C. An Alexa Fluor® 488-conjugated donkey anti-mouse IgG polyclonal (1/1000) was used as the secondary antibody.

  • The pictures show immunohistochemical staining of normal mucosa and neuroendocrine tumour of the small intestine.  The antibody (ab8049) stains positive in a 1:10 dilution in nerve cells of the colon as well as some cells in normal colon mucosa and some neuroendocrine tumors of the digestive tract. Staining protocol: Mouse monoclonal to Synaptophysin (ab8049), diluted 1:10 in PBS plus BSA, 60 min at room temperature,  
    secondary antibody: HRP solution with anti mouse polyclonal antibodies, 30min. DAB staining 10 min; Hematoxylin 30 sec.

  • Overlay histogram showing PC12 cells stained with ab8049 (red line). The cells were fixed with methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab8049, 1/20 dilution) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a decreased signal in PC12 cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween used under the same conditions.

Thank you for calling Abcam. The immunogen for ab8049 is the vesicular fraction of bovine brain and the epitope is to a pentapeptide repeat structure in the carboxy-terminal cytoplasmic tail of synaptophysin (Knaus & Betz 1990). Unfortunately the exact immunogen sequence is not known, and we are at this time unable to provide the peptide for ab8049. Based on this information I do not think that any of the synaptophysin peptides in our catalogue would be successful in blocking the activity of ab8049. There is always a chance though. Thank you for your enquiry. I am sorry to hear that you are experiencing difficulties with this product ab8049 in IHC. Often it is possible to make suggestions that help resolve problems. We will happily offer technical support and in the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 120 days of purchase), and if it appears that the antibody is at fault, a replacement/credit note/refund will be offered. Can you confirm that the second antibody you used also managed to produce good results with other primary antibodies? In some cases, the problem may be with the secondary antibody not working properly. Did you get any background staining when you run a “no primary” control? Most formalin-fixed tissue requires an antigen retrieval step before immunohistochemical staining can proceed. This is due to the formation of methylene bridges during fixation, which cross-link proteins and therefore mask antigenic sites. Antigen retrieval with Tris/EDTA pH 9.0 buffer is suitable for most antigens. Sodium citrate pH 6.0 is also widely used. Please use one of these buffers instead of deionised water. I am afraid that you are using this primary antibody (mouse host species) with mouse samples. This means that your secondary antibody (anti-mouse IgG) will bind to all the endogenous IgG in the mouse tissue and high non-specific background would occur. There are a few suggestions we can make to try and improve your results when doing mouse-on-mouse staining. However, please note that your success may vary and we cannot guarantee that you will see a reduction in non-specific staining. IHCWorld.com describes an additional blocking step using unconjugated fab fragment anti-mouse IgG to block endogenous IgG present in the tissue sample. Please see the following link for further information: http://ihcworld.com/_protocols/general_IHC/immuno_mom.htm In addition, kits such as Dako’s "Animal Research kit" (ARK) allows the direct biotinylation of the primary Ab being used, negating the necessity for a secondary Ab and reducing the potential for non specific staining.
