  • IHC image of ab45769 staining in human hippocampus formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab45769, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

A.In general abcam antibodies are fantastic (even though they are quite expensive for the amount the have), i have had now two antibodies from you that did not work at all and made me loose a lot of time and most importantly samples!. You should have a better quality control of batches, even if it is from your costumers

The antibody should detect a single band however the multiple bands problem could be due to protease inhibitors being not added, the lysis buffer not at 4C, high conc. of primary and secondary etc. In order to obtain the best results we always recommend customers to troubleshoot the protocol; tweaking few steps e.g. dilution factor or blocking always help to get rid of extra multiple bands. I am happy to provide the free of charge replacement. However we do have another antibody ab128690 which has quite a promising WB image, would you like to try this one.

Please be advised that there is no robust protocol for antibodies. The dilution factor always needs to be optimized because no 2 lysates or 2 labs conditions are the same. I would suggest trying a range between 1/500, 1/1000 and 1/2000 dilution with ab128690
