  • Relevance
    The majority of human adenocarcinomas including colorectal, pancreatic, gastric, ovarian, endometrial, mammary, and non-small cell lung cancer display some cell populations that are positive for TAG72. About 60% of carcinoma patients express TAG72 in their sera. TAG72 is reportedly useful in distinguishing pulmonary adenocarcinomas from pleural mesotheliomas.

  • ab16838 staining human colon carcinoma by IHC-P.
  • ICC/IF image of ab16838 stained MCF-7 cells. The cells were 4% formaldehyde (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab16838, undiluted) overnight at +4°C. The secondary antibody (green) was ab96879 Dylight 488 goat anti-mouse IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Both of these antibodies are tissue culture supernatant meaning that it is not possible to determine antibody concentration, since tissue culture supernatant contains proteins besides the antibody of interest. Supernatant is the fluid resulting from centrifugation of hybridomas in tissue culture that are secreting specific monoclonal antibodies. Supernatants therefore typically contain culture medium, 5-10% fetal calf serum, and other proteins. Antibody concentration is only relevant for purified antibodies.
