    • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

  • Target

    • Relevance

TCR (T cell antigen receptor) recognizes foreign antigens and translates such recognition events into intracellular signals that elicit a change in the cell from a dormant to an activated state. TCR is a heterodimer composed of either alpha and beta or gamma and delta chains. CD3 chains and the CD4 or CD8 coreceptors are also required for efficient signal transduction through the TCR.

        Predicted band size: 16 kDa
Observed band size: 21 kDa why is the actual band size different from the predicted?
Additional bands at: 56 kDa, 74 kDa. We are unsure as to the identity of these extra bands.

        Exposure time: 90 seconds

TCR alpha contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.

      Question

I am interesting in using your anti-TCRalpha polyclonal antibody, but I need to know if you have some information regarding its funcionality in FACS or as stimulating antibody. Also I would like to ask if you know the epitope in TCRalpha that is recognized by this antibody.

      Answer

Thank you for your enquiry. To our knowledge, the antibody has yet to be tested in these applications, and I am sorry this means I am unable to supply you with further details on FACS or its functionality as a stimulating antibody. All tested applications are specified on Abcam product datasheets. As this antibody is a polyclonal antibody to the whole human recombinant protein, it will recognise many epitopes on the protein. If you require more specificity on the epitope, you may like to try a monoclonal antibody. If you decide to go ahead and purchase this product for these new applications, I would be very pleased to hear how you get on . We would be very grateful if you could submit an Abreview via the online product datasheet. We always encourage customers to send their results back to us, whether positive or negative, and make all product information available to researchers. If you send us an Abreview, we can offer you from 50 up to 300 Abcam points (Abpoints) based on submission time and type of application. The less characterised the product you review, the more Abpoints you will receive. Furthermore, you get 100 extra Abpoints for submitting an image. These Abpoints can then be claimed back as rewards. I hope this information helps, please do not hesitate to contact us if you need any more advice or information.
