Target

  • Relevance

    Tetanus toxin acts by inhibiting neurotransmitter release. It binds to peripheral neuronal synapses, is internalized and moves by retrograde transport up the axon into the spinal cord where it can move between postsynaptic and presynaptic neurons. It inhibits neurotransmitter release by acting as a zinc endopeptidase that catalyzes the hydrolysis of the 76-Gln-|-Phe-77 bond of synaptobrevin-2. The precursor polypeptide is subsequently cleaved to yield subchains L and H. These remain linked by a disulfide bridge and are non-toxic after separation.

Question

I am going to do WB against recombinant universal T-helper cell epitope, callad tt830-844: a.a. qyikanskfigite. I think this three Ab (ab53829, ab68494 and ab35171) can work. Can you confirm if you used the whole protein to induce the imune response? If these are not suitable can you please recommend one.

The immunogen information is published on each of the product datasheets unless it is a commercially sensitive information. Could I advise you and your customer to read the on-line documents to check the tested applications as well.

ab35171: Synthetic peptide conjugated to KLH, corresponding to amino acids 1300-1314 of Tetanus Toxoid (Clostridium Tetani).

ab53829: Tetanus toxoid (formaldehyde inactivated tetanus toxin).

ab68494: Purified toxin.

These tree antibodies are polyclonals so they recognize multiple epitopes. Therefore, fo rpolyclonals epitop mappig is not performed. If the epitope sequence for monoclonal antibodies are mapped then the information is listed on the datasheets under the "Epitope" section.

I have a customer with the following questions: Item 1 - Ab53589 Goat polyclonal to Tetanus Toxin. - Does this antibody react with the entire toxin or just the C-fragment? Item 2. Ab53829 Rabbit polyclonal to Tetanus Toxin. - What concentration is recommended for WB and ELISA? (not listed on data sheet - but would it be similar to the ab53589?) - I realise they are different concentrations.

Thank you for your enquiry. I can confirm that ab53589 can detect the tetanus toxin which is produced by the soil micro-organism Clostridium tetani, and also the C-fragment of tetanus toxin. Meanwhile, for ab53829 ELISA and WB recommend starting dilution, please try 1/500. However, please bear in mind that the optimal dilutions/concentrations should be determined by the end user.
