    • ab36495 at 1/1000 staining mouse kidney tissue sections by IHC-P. The tissue was paraformaldehyde fixed and a heat mediated antigen retrieval step was perfomred in citrate buffer. The tissue was then blocked and incubated with the antibody for 1 hour. A biotinylated rabbit anti-mouse antibody was used as the secondary.

    • ab36495 staining GF beta 2 in Human MCF7 cells by Immunocytochemistry/ Immunofluorescence.

      Cells were fixed with formaldehyde, permeabilized with 0.1% Tween/TBS and blocked with 10% BSA for 30 minutes at room temperature. Samples were incubated with primary antibody (1/50 in 10% BSA/TBST) for 1 hour. An AlexaFluor®488-conjugated goat anti-mouse monoclonal IgG (1/250) was used as the secondary antibody.

    I learned from the lab is that the antibody was validated on purified recombinant TGF-beta 2, and there is no unusual protocol requirement for getting good western blot results.

    It is possible that your samples just have low amounts of the protein, below the detection limit of the antibody, but it does appear to be non-specific, in any case. I will be happy to send a replacement free of charge. I recommend a different antibody, for instance ab113670, which detects a band for tissue samples at about 55 kDa which we assume is TGF beta 2 plus the propeptide, with glycosylation. I can also issue a refund or credit, if you prefer.

    TGF beta 1 is often observed at about 12 kDa on blots, since the 48 kDa precursor is processed to a 12 kDa. I suspect that the band you observe between the two lower molecular weight markers is the 12 kDa subunit of the TGF beta homodimer, but this is just an assumption on my part. Can you please tell me the sizes of those molecular weight markers?

    This shows the subunit at about 12 kDa and the homodimer at 24 kDa.

    This antibody was tested in WB with recombinant TGFb2 which therefore can be used as positive control. We also have a positive Abreview from a customer which used mouse kidney and had veryclear positive staining.

    Other positive tissues according to the EST profile of TGFb2are: Thyroid, mammary gland, connective tissue, etc.

    ab9758 - the major band at44.3 kDa corresponding to the unprocessed (pre-pro-TGF beta 1) form, the mature processed TGF beta 1 is a 25 kD homodimer composed of two 12.5 kDa subunits joined by disulfide bonds. As the Western blot image indicates a 25 kDa band is seen in the non-reducing lane and a 12 kDa band in the reducing lane.
