TGF beta 1 is often observed at about 12 kDa on blots, since the 48 kDa precursor is processed to a 12 kDa. I suspect that the band you observe between the two lower molecular weight markers is the 12 kDa subunit of the TGF beta homodimer, but this is just an assumption on my part. Can you please tell me the sizes of those molecular weight markers?
For an example of a blot comparing reduced TGF beta 1 and unreduced homodimer, please see the blot on the datasheet of antibody ab9758:
This shows the subunit at about 12 kDa and the homodimer at 24 kDa.
