    Question

The antibodies ab59523 (Anti-NLN) and ab154173 (Anti-Thimet oligopeptidase) recognise two human peptidases of the M3 family that are rather similar at the sequence level (please find as attachment a protein sequence alignment).
I was wondering if you could tell me (based on your knowledge of the epitopes) if there is possibility of cross reaction in either case?
Also, if these particular antibodies would not be suitable to distinguish these two proteins, perhaps you could suggest others from your catalog.

This issue is very important for my research and will likely determine whether or not we will purchase these antibodies.

    Answer

I have researched the immunogens and alignments for you:

ab59523 Anti-NLN antibody : a blast of the RP3-Neurolysin antibody epitope shows about 59% identity with Thimet Oligopeptidase. It is immunogen affinity purified so there should be no or very little crossreactivity. This may also depend on the amount of TOP present in the sample.

ab154173 Anti-Thimet Oligopeptidase antibody [EPR7651]: similarly, the immunogen has 53% alignment to the human NLN protein. This one is not affinity purified.
