    The antibody recognizes the expected additional band corresponding to the ternary complex of human TS-dFUMP-reduced folate in HeLa cells treated with the TS inhibitor 5-FUdR. This event occurs in most human breast, colorectal, gastric, head and neck carcinomas. The antibody recognizes 36 kDa human TS.

  • ab7398 is shown to detect thymidylate synthase present in a HeLa cell extract. Each lane is estimated to contain 4 µg of protein. Lanes 1, 2 and 3 represent 1/2000, 1/5000 and 1/10000 fold dilutions of the antibody. Detection was made using HRP Rabbit-a-Sheep IgG diluted 1/1000 and color development using TMB substrate for approximately 4'.
  • IHC image of Thymidylate Synthase staining in Human colon carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab7398, 5µg/ml, for 15 mins at room temperature. A Rabbit anti-Sheep biotinylated secondary antibody was used to detect the primary, and visualized using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

All the cells especially the rapidly proliferating cells express this enzyme. This is a polyclonal antibody, and so we cannot refer to subclasses. The concentration is 10mg/ml and the sensitivity can be seen from the recommended concentrations for each application.
