Target

  • Function

    Plays an important role in the organization of the cytoskeleton (By similarity). Binds to and sequesters actin monomers (G actin) and therefore inhibits actin polymerization.
    Seraspenide inhibits the entry of hematopoeitic pluripotent stem cells into the S-phase.

    Expressed in several hemopoietic cell lines and lymphoid malignant cells. Decreased levels in myeloma cells.

Reviewing this case, I would like to offer some suggestions to help optimize the results between batches. I would also appreciate if you can confirm some further details:

1. I would like to reassure you that we monitor feedback closely on a weekly basis and we are not currently concerned about the general quality of this antibody or this batch. There have been no reported changes in results. However, there can be some batch variation with polyclonal antibodies. It may be that a lower concentration of antibody is required for this batch to decrease the background signal in your experiments. What dilutions have already been tried?

2. I am sorry to confirm that thi antibody is not tested and covered by the 6 month guarantee in IHC-P, However, as it has worked before for you we can make an exception and provide the guarantee in this case.

3. Antigen retrieval can sometimes require optimization. I can suggest to try different times, for example 5. 10, 20 and 30 minutes. Also, it may be useful to consider trying citrate buffer heat mediated retrieval.

4. I can recommend to include 0.2% Tween in the wash buffer. This will help to wash away ay excess antibody.

Question

Inquiry: Hello, We have been using this antibody for IHC on paraffin embedded samples and was working fine with the previous lot (944043). However, we have been experiencing problems with the new lot we have recently received (GR2453-5). Instead of getting the specific staining we usually see the whole tissue turns brown within seconds when developing with DAB. The negative controls are ok and we haven't changed anything with the protocol we were using before. I was wondering if something has changed between the two lots. Kind Regards

I would like to reassure you that we monitor feedback closely on a weekly basis and we are not currently concerned about the general quality of this antibody or this batch.

This antibody is tested and covered by our 6 month guarantee for ELISA and WB and in human samples. The antibody was tested against the immunizing peptide (Thymosin ß4 synthetic peptide) but you should also see high levels of the protein in thymus extract. Thank you for your enquiry regarding ab14334. I can confirm that the antibody has currently only been tested in ELISA on the human synthetic peptide used to generate this antibody. We do not currently supply this peptide or a positive control and I think that human thymus extract may be a good positive control to try. However should you wish to purchased the immunizing peptide please let me know and I will try to source this to add to our catalogue for you.
