    • Function

      mRNA-binding protein that plays a key role in self-renewal of erythroid cells in response to glucocorticoids. Specifically binds to the AU-rich element (ARE) in the 3'-UTR of target mRNAs, promoting their deadenylation and degradation (PubMed:14981510). Specifically expressed in burst-forming unit-erythroid (BFU-E) progenitors in response to glucocorticoids and acts as a negative regulator of erythroid cell differentiation: promotes self-renewal of erythroid cells by binding mRNAs that are induced or highly expressed during terminal erythroid differentiation and promotes their degradation, preventing erythroid cell differentiation. Down-regulated during erythroid differentiation from the BFU-E stage, stabilizing mRNAs required for terminal differentiation.
    • Involvement in disease

      Defects in ZFP36L2 may be a cause of leukemias. Frameshfits mutations disrupting ZFP36L2 have been found in a patient with acute myeloid leukemia (PubMed:21109922).

      I have asked for "sample size" antibody, if you have them available. The "100 ug" is a full-size vial which would suffice for at least a dozen of WB applications. We just would like to see how the Ab worked and if they would distinguish the isoforms (ZFP36, L1 and L2) before buying the full size.

      I have heard back from the lab regarding ab83579:


      Crossreactivity with TISIIB and TISIID isn't something we have tested in house but an immunogen search shows that both TISIIB and TISIID only have 4 residues of the immunogen in common so we would say it is very unlikely that ab83579 would detect these 2 proteins.
      We do not offer smaller samples sizes than our selling size (due to operational resons), but have a guarantee in place that covers the antibody for 6 months after the purchase for the use in the tested species and applications as listed on the datasheet (https://www.abcam.com/Abpromise).

      I have received the following information regading 2 of the antibodies. For ab83579, I am still waiting for more information.

      ab70775:
      The antibody was raised against human TIS11D. This antibody has been validated for Western blot using A549 cell extracts. This antibody has not been tested against TIS11A or TIS11B. Alignment of the antigen sequence with TIS11A (http://www.uniprot.org/uniprot/P2665) shows a 60% homology.
      Alignment of the antigen sequence with TIS11B (http://www.uniprot.org/uniprot/Q07352) shows a 66.7% homology.

      ab53747:
      We have not obtained data from endogenous protein and have not tested cross reactivity with TIS11A and TIS11D.
      However, we checked cross reactivity using “BLAST search” between immunogen sequence of ab53747 and TIS11A, TIS11D. And the results are the homology is for TIS11D: 76%, and for TIS11A: 68%. Crossreactivity might be possible for TIS11D.

      The immunogen sequence used to create this antibody is from aa231-270 ofhuman TIS11D. This sequence contains a 15aa span which has a 70% homology with TIS11B. At this level, we do not consider ab70775 to be cross reactive with TIS11B.

      Western blot is smaller than the band predicted by sequence. However, migration is affected by other factors and so the actual band size observed may differ from that predicted. Common factors include...

      post-translational modification- e.g. phosphorylation, glycosylation etc which increases the size of the protein
      post-translation cleavage- e.g. many proteins are synthesized as pro-proteins, and then cleaved to give the active form, e.g. pro-caspases
      splice variants- alternative splicing may create different sized proteins from the same gene
      relative charge- the composition of amino acids (charged vs non-charged)
