Inquiry: Order Details: Antibody code: ab104513 Batch number: GR28719-4 General Information: Antibody storage conditions (temperature/reconstitution etc) – sample was aliquoted into smaller volumes and stored in freezer (-20°C) Description of the problem (high background, low signal, non-specific staining etc.) – Non-specific (inappropriate) staining. We noticed color spots on some irrelevant areas of the slide. It seemed like we added some coloured not soluble Ab sample. Sample (Species/Tissue/Cell Type/Cell Line etc.). Human lung cancer paraffin embedded sections Fixation of sample (Ethanol/Methanol/Acetone/Paraformaldehyde/Other/Duration etc.). Formalin Antigen retrieval (Enzymatic method, Heat mediated technique etc.). Water bath 60 min 95°C in Zytomed Systems OmniPrep Solution Permeabilization step – we do not need it and didn’t perform Blocking conditions (Buffer/time period, Blocking agent etc.) Protein Block solution from the REVEAL Rabbit Specific AP Conjugate kit (Spring) (0.5% BSA, 0.5% casein) Primary Antibody (Diluent/Dilution/Incubation time, Wash step): aTF diluted 1:100 and 1:40 in the antibody diluent from the kit (PBS-0.1% BSA). Incubation at 4°C, o/n. Washing in 1XPBS/0.1% Tween-20 Secondary Antibody (Manufacturer/Species/Diluent/Dilution/Incubation time, Wash step). REVEAL Rabbit Specific AP Conjugate kit (Spring) – ready to use – 30 min at RT Detection method Naphhtol Phosphate + Fast Red Chromogen (kit) Positive and negative controls used (please specify).
