Can you tell me the percentage of monocytes that express TLR4 using this antibody in FACS on live cells? Can you provide me with a figure comparing a plot using the antibody versus an isotype control?

Thank you for your enquiry. Expression of TLR4 on normal resting monocytes is low and for this reason ab11227 is routinely QC'd on the U937 cell line which demonstrates a reasonable level of expression. I can attach an image here so I will send you a separate email with some histograms. However, I am pleased to say that I was able to obtain a histogram showing our current batch of ab11227 tested on monocytes. Expression of TLR4 is low, overlapping the isotype control, but still distinct. I believe that TLR4 expression on this cell type can be upregulated following stimulation with e.g. LPS or IFN gamma. You could refer to the literature for further information but the following paper will provide a good starting point. Bosisio, D. et al. (2002).
