The details you have kindly provided will enable us to investigate this case for you and also gives us vital information for our monitoring of product quality
I appreciate the time you have spent in the laboratory and understand your concerns. It is regrettable the results have not been successful. I would like to reassure you that this antibody is tested and covered by our guarantee for WB and mice.
Before deciding how to proceed with this particular case, I would like to offer some suggestions to help optimise the results. I would also appreciate if you can confirm some details of the procedure.
1. Could you please confirm how the knock out was achieved. In human we found an isoform 2 of the TMEM132D with a molecuar weight of 70 kDa. Furthermore in the WB image on our datasheet this 70 kDa band is detected in mouse liver lysate.
2. Could you confirm whether it was tested if the knock out is achieved for both isoforms? For example has this been tested with RT-PCR?
3. It would be beneficial to run a positive control of mouse liver lysate for this experiment if not already done. That would show if the antibody does not detect the 122 kDa protein in a tested sample.
4. If possible the amount of loaded protein should be between 20-30 ug. If the protein is expressed at a low level this amount of protein might be crucial for the detection. Because the used antibody is polyclonal the affinity against the protein can be vary for different lots. This is due to different immunisation rounds and hosts.
5. We would suggest it might be beneficial to reduce the dilution of primary antibody for the detection. We recommend a 1/500 dilution for WB with antibodies out of whole antiserum.
