The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Application | Abreviews | Notes |
---|---|---|
IHC-P | Use at an assay dependent concentration. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol. | |
WB | 1/1000. Detects a band of approximately 174 kDa (predicted molecular weight: 174 kDa). | |
ICC | 1/50 - 1/100. |
Target
-
Function
Control of topological states of DNA by transient breakage and subsequent rejoining of DNA strands. Topoisomerase II makes double-strand breaks. -
Sequence similarities
Belongs to the type II topoisomerase family. -
Post-translational
modificationsPhosphorylation has no effect on catalytic activity. -
Cellular localization
Cytoplasm. Nucleus > nucleoplasm. Generally located in the nucleoplasm. - Information by UniProt

Immunocytochemistry analysis of HeLa (human cervix adenocarcinoma epithelial cell) labeling Topoisomerase II alpha with purified ab52853 at 1/50 dilution (4 µg/ml). Cells were fixed with 4% Paraformaldehyde and permeabilised with 0.1% tritonX-100. Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) at 1/1000 (2 µg/ml) was used as the secondary antibody. ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1/200 (2.14 µg/ml) was used as counterstain. Nuclei were stained blue with DAPI.
Negative control: PBS instead of the primary antibody.
