RelevanceTPO acts as an enzyme and plays a central role in thyroid gland function. The protein functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Multiple transcript variants encoding distinct isoforms have been identified for this gene. Additional splice variants have been described but their biological natures have not been determined.

Western blot analysis of varying levels of Recombinant human TPO protein using ab271257 at 0.1 µg/ml.
Used in conjunction with compatible secondary reagents the detection limit for recombinant TPO is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
Human TPO was detected by sandwich ELISA (using 100 µl/well) using a concentration of 0.25-1.0 µg/ml of ab271257. This antibody in conjunction with an appropriate capture antibody, allows the detection of at least 0.2 – 0.4 ng/well of recombinant TPO protein.
