  • Function

    Lysosomal serine protease with tripeptidyl-peptidase I activity. May act as a non-specific lysosomal peptidase which generates tripeptides from the breakdown products produced by lysosomal proteinases. Requires substrates with an unsubstituted N-terminus.

    Detected in all tissues examined with highest levels in heart and placenta and relatively similar levels in other tissues.
  • Involvement in disease

    Defects in TPP1 are the cause of neuronal ceroid lipofuscinosis type 2 (CLN2) [MIM:204500]. A form of neuronal ceroid lipofuscinosis. Neuronal ceroid lipofuscinoses are progressive neurodegenerative, lysosomal storage diseases characterized by intracellular accumulation of autofluorescent liposomal material, and clinically by seizures, dementia, visual loss, and/or cerebral atrophy. The lipopigment pattern seen most often in CLN2 consists of curvilinear profiles.

The TTP1 antibody ab88305 was generated against full-length human protein, not a peptide. Alignment of the human and canine TTP1 shows 90% identity, using the sequences at these UniProt pages,

Human http://www.uniprot.org/uniprot/O14773.

The antibody is polyclonal, meaning it is a collection of antibodies that recognize different epitope sequences on the TTP1 protein. Most are expected to be identical in human and canine, given the 90% overall identity. However, it has not been tested for reactivity with canine samples by IHC or any other applications. That is also the case for our other TTP1 antibodies: untested against canine samples.

I suggest also considering the rabbit monoclonal ab195234, a more recent addition to our catalogue. It is raised against a fragment of human TTP1 which is 91% identical to the homologous canine sequence.
