Application Abreviews Notes
IHC-P Use a concentration of 20 µg/ml.
WB 1/500. Detects a band of approximately 32 kDa.Can be blocked with TRAIL peptide (ab7867).

Gives a 32 kD band which represents the membrane-bound form of TRAIL.

  • Function

Cytokine that binds to TNFRSF10A/TRAILR1, TNFRSF10B/TRAILR2, TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and possibly also to TNFRSF11B/OPG. Induces apoptosis. Its activity may be modulated by binding to the decoy receptors TNFRSF10C/TRAILR3, TNFRSF10D/TRAILR4 and TNFRSF11B/OPG that cannot induce apoptosis.
  • Tissue specificity

Widespread; most predominant in spleen, lung and prostate.
  • Sequence similarities

Belongs to the tumor necrosis factor family.

Thank you for your enquiry. Ab2056 recognizes both the whole TRAIL protein (32 kDa) as well as its extracellular domain (19 kDa). If you are running a Western with HeLa whole cell lysate (which is recommended for a positive control) you would expect to see a band at 32 kDa. I will email you an image separately. If you are running recombinant TRAIL protein, you would see a band at 24 kDa - Recombinant human TRAIL monomer has a molecular mass of approximately 21 kDa. Due to glycosylation, recombinant human TRAIL migrates as a 24 kDa protein in SDS PAGE. The originator of this antibody used the following for the lysis buffer: HeLa cell lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylenediaminetetraacetic acid, 1 mM phenylmethylsulfonyl fluoride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecylsulfate, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The cell lysate was boiled for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol. If you have any additional questions, please contact us again.
