Thank you for your enquiry. Ab2056 recognizes both the whole TRAIL protein (32 kDa) as well as its extracellular domain (19 kDa). If you are running a Western with HeLa whole cell lysate (which is recommended for a positive control) you would expect to see a band at 32 kDa. I will email you an image separately. If you are running recombinant TRAIL protein, you would see a band at 24 kDa - Recombinant human TRAIL monomer has a molecular mass of approximately 21 kDa. Due to glycosylation, recombinant human TRAIL migrates as a 24 kDa protein in SDS PAGE. The originator of this antibody used the following for the lysis buffer: HeLa cell lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylenediaminetetraacetic acid, 1 mM phenylmethylsulfonyl fluoride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecylsulfate, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The cell lysate was boiled for 5 min in 1 x SDS sample buffer (50 mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol.
