Application Abreviews Notes
WB Use a concentration of 0.3 - 1 µg/ml. Detects a band of approximately 75 kDa (predicted molecular weight: 76 kDa).
IHC-P Use a concentration of 3 - 4 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Target

  • Relevance

    RNF139 is a multi-membrane spanning protein E3 ubiquitin ligase which is mutated in clear-cell renal carcinomas. It contains a sterol-sensing domain and a RING-H2 motif essential for ubiquitin ligase activity. It is a potential tumor suppressor for renal cell carcinoma. Human kidney cells are growth inhibited by RNF139. Inhibition is manifested by G2/M arrest and is dependent upon the ubiquitin ligase activity of the RING domain. Expression of RNF139 represses genes involved in cholesterol and fatty acid biosynthesis that are transcriptionally regulated by the sterol response element binding proteins (SREBPs). RNF139 modulation of SREBP activity comprises a novel regulatory link between growth control and the cholesterol/lipid homeostasis pathway.
