  • Function

    Receptor for brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-4/5 but not nerve growth factor (NGF). Involved in the development and/or maintenance of the nervous system. This is a tyrosine-protein kinase receptor. Known substrates for the TRK receptors are SHC1, PI-3 kinase, and PLC-gamma-1.
  • Tissue specificity

    Isoform TrkB is widely expressed, mainly in the nervous tissue. In the CNS, expression is observed in the cerebral cortex, hippocampus, thalamus, choroid plexus, granular layer of the cerebellum, brain stem, and spinal cord. In the peripheral nervous system, it is expressed in many cranial ganglia, the ophtalmic nerve, the vestibular system, multiple facial structures, the submaxillary glands, and dorsal root ganglia. Isoform TrkB-T1 is expressed in multiple tissues, mainly in brain, pancreas, kidney and heart. Isoform TrkB-T-Shc is predominantly expressed in brain.

  • ab6180 staining rat lung tissue sections by IHC-P.  Sections were fixed in acetone-methanol (3:2) and subjected to a heat mediated antigen retrieval step in citrate buffer (pH 6) prior to a 5 minute permeabilization in 0.5% TritonX-100 in PBS and blocking in 5% milk for 30 minutes at 20°C.  The primary antibody was diluted 1/100 and incubated with sample for 1 hour at 20°C.  A HRP conjugated donkey anti-rabbit antibody, diluted 1/50, was used as the secondary.

"We know of no reason why a 20kD band should be present. While it is possible that this might be a fragment of the intact molecule, there is nothing that we no of in the literature that would explain its presence.
Our only suggestion to your customer is to ensure that the tissue is well protected with protease inhibitors during the homogenization/ extraction procedure to avoid proteolytic cleavage of the TrkB during processing. "

The tissue that we use as a positive control is the rat dorsal root ganglion. Using PFA containing picric acid as the fixative a population of neurons with medium to large diameter are stained. This population has been well described in the literature as being trkB positive. Paraffin sections have not been tested, but it works well in frozen sections. A blocking peptide is available for this antibody.

PFA alone or with picric acid will both give good staining, although we prefer the addtion of picric acid. Pre-perfusion is with sodium nitrite (1% in phosphate buffer). In rat dorsal root ganglia, the neurons stained are the in the medium to large range as has been published by several other workers. This antibody appears to have the same characteristics as those described in Yan et al (1997) J Comp Neurol 378: 135-157.
