  • Function

    Troponin I is the inhibitory subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity.
  • Involvement in disease

    Defects in TNNI2 are a cause of distal arthrogryposis type 2B (DA2B) [MIM:601680]; also known as arthrogryposis multiplex congenita, distal, type 2B (AMCD2B). DA2B is a form of inherited multiple congenital contractures. Affected individuals have vertical talus, ulnar deviation in the hands, severe camptodactyly, and a distinctive face characterized by a triangular shape, prominent nasolabial folds, small mouth and a prominent chin.

I have confirmed that the immunogen for ab126824 is full-length recombinant protein, not a fragment. The epitope has not been mapped. The trouble you are having with the sandwich ELISA may be a consequence of the two antibodies recognizing overlapping epitopes but it is more likely that one of the antibodies is failing. If you can, try checking for this with an indirect ELISA against antigen coated directly to the plate, followed by blocking with BSA, the antibody, and a secondary conjugated antibody.

For the sandwich, I recommend capturing with the monoclonal and detecting with the polyclonal ab123876, assuming you have an anti-rabbit secondary. Ideally, it should be absorbed to remove reactivity with mouse IgG1, the isotype of ab126824.
