Target

  • Function

    Receptor-activated non-selective cation channel involved in detection of pain and possibly also in cold perception and inner ear function. Has a central role in the pain response to endogenous inflammatory mediators and to a diverse array of volatile irritants, such as mustard oil, garlic and acrolein, an irritant from tears gas and vehicule exhaust fumes. Acts also as a ionotropic cannabinoid receptor by being activated by delta(9)-tetrahydrocannabinol (THC), the psychoactive component of marijuana. Not involved in menthol sensation. May be a component for the mechanosensitive transduction channel of hair cells in inner ear, thereby participating in the perception of sounds. Probably operated by a phosphatidylinositol second messenger system.

    Sections were incubated with primary antibody (1/1000) overnight at 4°C. An AlexaFluor®488-conjugated anti-rabbit IgG was used as the secondary antibody. ab36001 was used to stain CGRP (red).

  • Immunohistochemical analysis of wholemount murine colonic wall, staining TRPA1/TSA with ab58844.

    Wholemount preparations were incubated with primary antibody (1/1000) overnight at 37°C. An AlexaFluor®488-conjugated anti-rabbit IgG was used as the secondary antibody.

I can confirm thatab58844 Anti-TRPA1 antibodyis sold as whole antiserum. Unpurified antibodies, such as those sold as whole antiserum, ascites or tissue culture supernatant will not have a concentration stated on the datasheet. Antibody concentration is usually determined by protein assay, and serum will contain a lot of other proteins, which means the antibody quantification would not be accurate.

Generally we know that for whole antiserum, concentration of antibody is known to vary between 1 - 10 mg/ml.

We have two reports from customers who used the antibody ab58844 for western blotting. In both cases, the antibody was incubated with the membrane at a dilution of 1/1000.

The sizes of the bands detected by each customer differed. (I am assuming you are interested in the size of TRPA1, and not the antibody itself, which, denatured and reduced, consists of two 50 kDa heavy chains and two 25 kDa light chains).

One customer detected a band of about 80 kDa using human cells. The customer notes, "Although accordingly to the literature TRPA1 is a protein of circa 127 kDa we have found a specific band at circa 75 KDa and this has been also reported in another work using the same cells (keratinocytes) (Journal of Investigative Dermatology (2009) 129, 2312–2315 doi:10.1038/jid.2009.58 published online 12 March 2009)."

The other customer saw a specific band between 90-100 kDa using mouse tissue, which suggests that the size of the protein detected in blots may be sample and/or species specific.

I apologize, but I misunderstood your original email and had assumed that you were having trouble seeing bands in WB. I didn't realize you were asking about the expected MW.
Anyway, this antibody has been shown by customers to recognize a band ˜80 kDa using human cells. The customer notes, "Although accordingly to the literature TRPA1 is a protein of circa 127 kDa we have found a specific band at circa 75 KDa and this has been also reported in another work using the same cells (keratinocytes) (Journal of Investigative Dermatology (2009) 129, 2312–2315 doi:10.1038/jid.2009.58 published online 12 March 2009)."
