  • Function

    Receptor-activated non-selective cation channel involved in detection of pain and possibly also in cold perception and inner ear function. Has a central role in the pain response to endogenous inflammatory mediators and to a diverse array of volatile irritants, such as mustard oil, garlic and acrolein, an irritant from tears gas and vehicule exhaust fumes. Acts also as a ionotropic cannabinoid receptor by being activated by delta(9)-tetrahydrocannabinol (THC), the psychoactive component of marijuana. Not involved in menthol sensation. May be a component for the mechanosensitive transduction channel of hair cells in inner ear, thereby participating in the perception of sounds. Probably operated by a phosphatidylinositol second messenger system.

  • ab62053 staining TRPA1/TSA in murine skin tissue by Immunohistochemistry (Frozen sections).Tissue was fixed in methanol, permeabilized with 0.4% Triton X-100, blocked using 10% serum for 30 minutes at 25°C and then incubated with ab62053 at a 1/100 dilution for 16 hours at 4°C. The secondary used was an Alexa-Fluor 594 conjugated goat anti-rabbit polyclonal used at a 1/500 dilution.

I have tried the TRPA1 antibody (ab62053) for a few times. It didn't produce any staining on human and mice intestine frozen sections. When I applied it on mouse DRG cell culture, it seemed to stain everything (see attached images). It has been estimated that only a small proportion of DRG expressing TRPA1. Below is my protocol. Do you have any suggestions? Thanks.

Thank you for contacting us. I am very sorry to hear that also with the ab62053 the results are not satisfying. I have looked at the protocol and the only thing I can suggest, is to try also the Methanol fixation. Indeed, the result on the datasheet has been obtained with Methanol fixation. Methanol and paraformaldehyde fixation are very different and can results in different availability of the epitopes. However as this antibody has also been tested on paraffin embedded, PFA fixed sections, I would expect it also to work on PFA fixed frozen sections- although there was no antigen retrieval.
