    TRRAP (TRansformation/tRanscription domain Associated Protein) is an adapter protein that is found in a number of multiprotein chromatin complexes with histone acetyltransferase (HAT) activity. These complexes include PCAF, TFTC-HAT, TIP60 HAT, STAGA, and BAF53 complexes. TRRAP is thought to be responsible for the concerted and context-dependent recruitment of these complexes to chromatin during transcription, replication and DNA repair. TRRAP plays a key role in transcriptional activation by c-Myc, p53/TP53, E2F1 and E2F4.

  • Cross-linked ChIP was performed with HeLa (human epithelial cell line from cervix adenocarcinoma) chromatin extract and 5 μg of either control rabbit IgG or ab227590. The precipitated DNA was detected by PCR with primer set targeting to nucleolin promoter.

  • TRRAP was immunoprecipitated from HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell extract with 4 µg of ab227590. Western blot was performed from the immunoprecipitate using ab227590 at 1/1000 dilution. Anti-rabbit IgG was used as a secondary reagent.

    Lane 1: 4 μg of preimmune Rabbit IgG instead of ab227590 in HEK-293T whole cell extract.
    Lane 2: ab227590 IP in HEK-293T whole cell extract.

    5% SDS-PAGE gel.
