Immunofluorescent analysis of HeLa (human epithelial cell line from cervix adenocarcinoma) cells stained for TTC11/FIS1 (green) using ab229969 at 1/100 dilution in ICC/IF, followed by Alexa Fluor 488® congugated Goat Anti-Rabbit IgG (H+L).
Paraffin-embedded human kidney tissue stained for TTC11/FIS1 using ab229969 at 1/600 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
TTC11/FIS1 was immunoprecipitated from 500 µg MCF7 (human breast adenocarcinoma cell line) whole cell lysate with 4 µg of ab229969.
Lane 1: Rabbit monoclonal IgG (1 µg) IP instead of ab229969 in MCF7 whole cell lysate.
Lane 2: ab229969 IP in MCF whole cell lysate.
Lane 3: MCF7 whole cell lysate 20 ug (input).
For western blotting, an HRP-conjugated light chain specific antibody was used as the Secondary antibody at 1/50000 dilution.
