Thank you for these additional details. I would recommend increasing your incubation with primary antibody to overnight at 4 degrees to get the strongest signal. This is how we routinely test our antibodies for IHC. You may also wish to reduce the antigen retrieval time, or do several time points testing at 20, 30 and 40 minutes to get the optimal retrieval. Please let me know if these suggestions do not improve your results.
