  • Immunohistochemical analysis of TWEAKR/FN14 expression in paraffin embedded formalin fixed liver using 1/100 ab85089. Note membrane staining.

  • ICC/IF image of ab85089 stained HepG2 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab85089, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Do you still have problems with emailing technical@abcam.com? It might be worthwile to try one more time. I took a look at the images you sent: Yes, there seems to be some nuclear staining as well as nice plasma membrane staining. My suggestions would be to dilute the primary antibody further as the nuclear staining might be due to too much primary antibody. In addition or as alternative, using less secondary antibody might help as well. Further, increasing the blocking time could help improve the results, too. Should none of this have the desired effect, it might be best to try a different TWEAKR antibody, such as ab109365. Please let me know if these suggestions are of help. If not, please let me know if trying another antibody would help you. Further options would be a credit or refund. I hope this information is helpful to you.
