  • Function

    Involved in mitochondrial DNA (mtDNA) metabolism. Could function as an adenine nucleotide-dependent DNA helicase. Function inferred to be critical for lifetime maintenance of mtDNA integrity. In vitro, forms in combination with POLG, a processive replication machinery, which can use double-stranded DNA (dsDNA) as template to synthesize single-stranded DNA (ssDNA) molecules. May be a key regulator of mtDNA copy number in mammals.
  • Tissue specificity

    High relative levels in skeletal muscle, testis and pancreas. Lower levels of expression in the heart, brain, placenta, lung, liver, kidney, spleen, thymus, prostate, ovary, small intestine, colon and leukocytes. Expression is coregulated with MRPL43.

  • Western blot analysis of HeLa cell lysate labeling Twinkle with ab83329 at 1.0µg/ml.

    12% gel concentration
  • ICC/IF image of ab83329 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab83329, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

The human protein has 3 isoforms: 77, 66, and 60 kDa (http://www.uniprot.org/uniprot/Q96RR1).
Thus, the band you are seeing around 57 kDa could be the third isoform at about 60 kDa. Please keep in mind that the MW marker are not precise and a difference by 2 or 3 kDa is possible.

The bands could vary from sample type to sample type - depending on which isoform is present, and under which conditions the isoforms exist.

I assume you are using human samples?For another species, the results might vary as well.
