    - First try to dissolve a small amount of peptide in either water or buffer. The more charged residues on a peptide, the more soluble it is in aqueous solutions.
    - If the peptide doesn’t dissolve try an organic solvent e.g. DMSO, then dilute using water or buffer.
    - Consider that any solvent used must be compatible with your assay. If a peptide does not dissolve and you need to recover it, lyophilise to remove the solvent.
    - Gentle warming and sonication can effectively aid peptide solubilisation. If the solution is cloudy or has gelled the peptide may be in suspension rather than solubilised.
    - Peptides containing cysteine are easily oxidised, so should be prepared in solution just prior to use.

    UCP are mitochondrial transporter proteins that create proton leaks across the inner mitochondrial membrane, thus uncoupling oxidative phosphorylation from ATP synthesis. As a result, energy is dissipated in the form of heat.

Western blot
I had bought the ucp2 antibody (Ab97931) fro WB.
According to the online information on this antibody (From your website) as well as published literature, it seems that this protein should be 33-34Kda and has been shown to work in human cells and is predicted but not proven to work in mouse cell lines.
I have used this for mouse fibroblats and instead of getting the band at 33kda, I have got a very clear and convincing band at 25kDa (that is the nearest band for 33kDa). So I tried the blocking UCp-2 peptide to check the authenticity of this band.(ab107707). We ran the same samples in duplicates on the same gel. Once the protein had been transferred, the membrane was cut into two parts, with the same two samples on them. We used the primary antibody (ab97931) on one of the membranes and the other piece we incubated with the ucp2 peptide plus the antibody (mentioned above). As can be seen from the image, the bands completely disappeared when incubated with the ucp2 peptide, hence proving that the band were specific to the ucp2 antigen.
