Applications

Application Abreviews Notes
WB 1/1000. Detects a band of approximately 10 kDa (predicted molecular weight: 10 kDa).

We don’t recommend this antibody for rat in WB. In our hands rat tissues showed negative staining.

IHC-P 1/4000. Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

In IHC, under our experimental conditions, this antibody showed specific staining in bronchial epithelium cells of both rat and mouse; however, there was non-specific staining on the pulmonary alveoli of mouse lung. Our collaborator observed the correct staining on the airway epithelium of mouse lung using immunofluorescence detection method.

IHC-Fr 1/500.

(Use 1/100 for rat tissue).

Antigen retrieval: Heated citrate solution (10mM citrate pH 6.0 + 0.05% Tween-20).

IP 1/30.

We don't recommend this antibody for rat in IP.  In our hands rat tissues showed negative staining.

  • Immunohistochemical analysis of paraffin-embedded mouse lung tissue labeling Uteroglobin with ab213203 at 1/4000 dilution, followed by Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/500 dilution.

    Strong cytoplasmic staining on the airway epithelium of mouse lung. However, there is some non-specific cytoplasmic staining on the alveolar cells of mouse lung. The correct staining on the airway epithelium of mouse lung is observed when using immunofluorescence detection method.

  • Uteroglobin was immunoprecipitated from 0.35 mg of mouse lung lysate with ab213203 at 1/30 dilution.

    Western blot was performed from the immunoprecipitate using ab213203 at 1/1000 dilution.

    VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10,000 dilution

    Lane 1: Mouse lung lysate 10ug (Input).

    Lane 2: ab213203 IP in mouse lung lysate.

    Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab213203 in mouse lung lysate.

    Blocking and dilution buffer and concentration: 5% NFDM/TBST.

    Exposure time: 10 seconds.
