Target

  • Function

Acts as a transcriptional coactivator of ATF2.
  • Domain

The leucine-zipper domain is required for coactivation activity. When this domain is deleted, the protein is able to stimulate transcription from a number of gene promoters.
  • Post-translational
    modifications

    Phosphorylated.
  • Cellular localization

    Nucleus.

    • Form

UTF1 is a transcriptional coactivator expressed in pluripotent embryonic stem cells and extra-embryonic cells. Upon activation by Oct-3/4, UTF1 may be responsible for the rapid proliferation and tumorigenic properties of embryonic stem cells. Sox2 is also part of the Oct-3/4-containing complex able to bind the regulatory region of UTF1. UTF1 acts as a transcriptional coactivator of ATF2 and it binds to the N-terminal region of ATF2. Associates with the TFIID complex through interaction with TBP.

ICC/IF image of ab105090 stained mouse embryonic stem cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab105090, 1µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM. This antibody also gave a positive result in 4% formaldehyde fixed (10 min) mouse ES cells at 1µg/ml.
