A 1:100,000 dilution of the antibody gave an O.D.=1.0 in a 15 minute reaction using HRP conjugated goat anti chicken IgY (IgG) and TMB as the substrate. Appropriate specificity controls were run.

  • Relevance
    The V5 epitope tag is derived from a small epitope (Pk) present on the P and V proteins of the paramyxovirus of simian virus 5 (SV5). The V5 tag is usually used with all 14 amino acids (GKPIPNPLLGLDST), although it has also been used with a shorter 9 amino acid sequence (IPNPLLGLD).

  • ab9113 staining human fibrosarcoma cells (HT1080) by ICC/IF. Cells were transfected with V5- tagged transmembrane protein and PFA fixed without undergoing permeabilization and blocking steps. The primary antibody was used at 10µg/ml and incubated with the sample for 1 hour at 20°C. An Alexa Fluor® 546 conjugated donkey polyclonal anti-chicken (H+L) antibody, diluted 1/500, was used as the secondary.

I can confirm that ab24740 Myc tag antibody and ab9113 V5 tag antibody will be suitable for WB and ICC-IF experiments.

As you will be doing duel staining, you will need to use primary antibodies from different species, which means would need to order different secondary antibodies for their detection, and also secondary antibodies with different conjugates for these two applications.

Thank you for your response. Some researchers add the cryoprotectant glycerol to a final concentration of 50% to prevent freeze/thaw damage; glycerol will lower the freezing point to below -20oC. While this may be acceptable for many antibodies, only a small percentage of the antibodies we offer have been tested for stability in this storage condition and our guarantee only applies to antibodies stored as recommended on the datasheet. Storing solutions containing glycerol at -80oC is not advised since this is below the freezing point of glycerol. Please be aware that glycerol can be contaminated with bacteria. Since the storage buffer does not contain any cryoprotective agent, lower temperature than 4ᵒC may have impact on its activity.
