  • Relevance
    The V5 epitope tag is derived from a small epitope (Pk) present on the P and V proteins of the paramyxovirus of simian virus 5 (SV5). The V5 tag is usually used with all 14 amino acids (GKPIPNPLLGLDST), although it has also been used with a shorter 9 amino acid sequence (IPNPLLGLD).

Please be also advised that ab1274, ab9116, ab9134, ab84164 are the antibodies against tags. These tags are not naturally present in any cells. These products can only be used in detecting the tag conjugated recombinant proteins that are expressed in tissues or cells. I hope this information is helpful to you. Thank you for contacting us. These antibodies are against tags so species is not applicable for these products. "10 min in 4% paraformaldehyde and proceeds with the staining" is not enough information that we need for any recommendations. Please ask the customer to provide following info - Are they planning to use tissue sections or cells? What type of tissue section or cells they will be using e.g. Eye, Brain etc. - Are they planning Immunohistochemistry on frozen sections or paraformaldehyde sections thereby incubating sections with acetone or ethanol or treating sections with antigen retrieval method? - Now these antibodies are against tags so you may ask how they are expressing the tag in cells or tissues. Please provide the above information then only we can provide further confirmation.

Thank you for your patience. I have heard back from the originator of this antibody and your protocol looks fine, especially since the monoclonal worked under the same conditions. There may have been a problem with the vial/lot that you received and I can certainly offer you a different batch to try, an alternative antibody (I know that you mentioned the chicken polyclonal), or a refund. We don't have have any data to this point as to how ab1274 will work in immunofluorescence.
