  • Relevance
    The Vav family are Rho/Rac guanosine nucleotide exchange factors (GEFs), consisting of three members in mammalian cells (Vav, Vav2, Vav3) and one in nematodes (CelVav). First discovered based on its transforming properties, Vav is expressed mainly in hematopoietic cells and a few non-hematopoietic tissues, such as the pancreas and tooth enamels. As a signalling transducer, Vav is involved in T-cell activated transduction of T-cell antigen receptor (TCR). T-cell stimulated and tyrosine phosphorylated Vav acts as a catalyst in the exchange of guanosine nucleotides on Rac-1, a GTP binding protein. Using a mouse model, Vav expression has been determined to play an essential role in the cyctosketetal, proliferative, and apoptotic pathways for developing lymphoid cells and its signal response.

I would not worry about results where the observed molecular weights are 10 or even 15% above the expected molecular weights. As explained in our https://www.abcam.com/protocols/western-blot-faq, this is very common and explained by the post-translational modifications, such as phosphorylation, which increase the size of the proteins.

The difference between these two products is that ab76225 is a rabbit monoclonal antibody while ab47282 is a rabbit polyclonal antibody. Both of these antibodies were raised against the corresponding residues around the phophorylated site at the 174 amino acid in the Vav protein sequence. Unfortunately neither of these antibodies have been tested in flow cytometry experiments, thus we do not know if they will work or not. Either one of these antibodies is likely to work in flow cytometery since they are recognizing the proper tertiary structure of the protein as evidenced by the fact that they work in immunocytochemistry experiments.
