    • Function
      Growth factor active in angiogenesis, vasculogenesis and endothelial cell growth. Induces endothelial cell proliferation, promotes cell migration, inhibits apoptosis and induces permeabilization of blood vessels. Binds to the FLT1/VEGFR1 and KDR/VEGFR2 receptors, heparan sulfate and heparin. NRP1/Neuropilin-1 binds isoforms VEGF-165 and VEGF-145. Isoform VEGF165B binds to KDR but does not activate downstream signaling pathways, does not activate angiogenesis and inhibits tumor growth.
    • Tissue specificity
      Isoform VEGF189, isoform VEGF165 and isoform VEGF121 are widely expressed. Isoform VEGF206 and isoform VEGF145 are not widely expressed.
    • Involvement in disease
      Defects in VEGFA are a cause of susceptibility to microvascular complications of diabetes type 1 (MVCD1) [MIM:603933]. These are pathological conditions that develop in numerous tissues and organs as a consequence of diabetes mellitus. They include diabetic retinopathy, diabetic nephropathy leading to end-stage renal disease, and diabetic neuropathy. Diabetic retinopathy remains the major cause of new-onset blindness among diabetic adults. It is characterized by vascular permeability and increased tissue ischemia and angiogenesis.

      Belongs to the PDGF/VEGF growth factor family.
    • Cellular localization
      Secreted. VEGF121 is acidic and freely secreted. VEGF165 is more basic, has heparin-binding properties and, although a signicant proportion remains cell-associated, most is freely secreted. VEGF189 is very basic, it is cell-associated after secretion and is bound avidly by heparin and the extracellular matrix, although it may be released as a soluble form by heparin, heparinase or plasmin.

    In general we recommend to use EDTA pH9. This antigen retrieval method is optimal for the most antigens. Please see the attachment and this publication (Emoto et al., J Histochem Cytochem. 2005 Nov ;53 (11) : 1311-21.) for more information.

    ab28775 was used successfully by our customers without, with citrate buffer pH6 and with Tris pH10. Please see Abreviews on the datasheet.

    The image on the datasheetfor ab32512 was generated with citrate buffer pH6.

    The optimal protocol usually has to be determined by the end user individually.

    In regards to the negative control in IHC for VEGF staining, this will be rather difficult as VEGF is secreted and present in many tissues andblood vessels.     I'm sorry to hear you are having a problem with ab28775. Please see the antibody datasheet. The recommended dilution for this antibody is 1/50. Since you are using this at a 20-fold higher dilution, I believe this is the problem. Please try the antibody at 1/50 and let me know if the results are better. In addition, I would recommend incubating overnight at 4C since this will give a slower but more specific binding. We do not recommend staining with antibodies raised in the same species as that of your sample, since the secondary anti-mouse antibody will recognize the endogenous IgGs in the sample and may lead to high background. You may wish to use a mouse-on-mouse kit to try to lower the background, but we cannot guarantee that the staining will be successful. Pancreas or kidney can be used as positive controls.
