    I am potentially interested in purchasing anti-villin Ab [erp3490] (ab109516, Epitomic catalog no. 3722-1) for flow cytometry. According to your specs, it reacts with mouse villin, but you only include human data on your specs sheet. Could you send me flow and western data on mouse cells/tissue that support this claim, please? Also, why on western does the observed band, 95 kd, differ from the predicted band, 93 kd?

    Thank you for contacting us.

    With many products cross reactivity with mouse and rat is determined by western blot only (this is the case for ab109516). If we have only determined the cross reactivity via WB, we will state this information on the website and datasheet in the "Species Reactivity" section. I have attached an image of our WB data with mouse and rat for ab109516. We recommend the antibody for FC based on the fact that it worked for human samples and the antibody appears to be mouse and rat reactive due to our WB results.

    This product has an expected molecular weight of 93 kDa. In the image on the datasheet we do show that this product in near its expected size at around 95 kDa. There are many reasons why a protein may appear to be at a different size than expected.

    Markers especially those which are prestained are approximate values. It is difficult to determining precise size based on published weights for calibration markers. Even common proteins may have several slightly different size estimates reported in the literature, depending upon the methods of molecular weight measurement.

    Other reasons why a protein may run at an unexpected size include gel type and protein charge. Post translation modifications, cleavage, splice variants and multimers can affect some protein targets as well.

    Most proteins will give estimates within a few percent of their actual weight when compared to appropriate calibration markers. If prepared in the same way each time a protein should run reproducibly on a given gel type.
