Target

  • Function

    Vimentins are class-III intermediate filaments found in various non-epithelial cells, especially mesenchymal cells. Vimentin is attached to the nucleus, endoplasmic reticulum, and mitochondria, either laterally or terminally.
    Involved with LARP6 in the stabilization of type I collagen mRNAs for CO1A1 and CO1A2.
  • Tissue specificity

    Highly expressed in fibroblasts, some expression in T- and B-lymphocytes, and little or no expression in Burkitt's lymphoma cell lines. Expressed in many hormone-independent mammary carcinoma cell lines.

  • Domain

    The central alpha-helical coiled-coil rod region mediates elementary homodimerization.
    The [IL]-x-C-x-x-[DE] motif is a proposed target motif for cysteine S-nitrosylation mediated by the iNOS-S100A8/A9 transnitrosylase complex.
  • Post-translational
    modifications

    Filament disassembly during mitosis is promoted by phosphorylation at Ser-55 as well as by nestin (By similarity). One of the most prominent phosphoproteins in various cells of mesenchymal origin. Phosphorylation is enhanced during cell division, at which time vimentin filaments are significantly reorganized. Phosphorylation by PKN1 inhibits the formation of filaments. Phosphorylated at Ser-56 by CDK5 during neutrophil secretion in the cytoplasm. Phosphorylated by STK33.
    O-glycosylated during cytokinesis at sites identical or close to phosphorylation sites, this interferes with the phosphorylation status.
    S-nitrosylation is induced by interferon-gamma and oxidatively-modified low-densitity lipoprotein (LDL(ox)) possibly implicating the iNOS-S100A8/9 transnitrosylase complex.

Question

I’m looking to characterise a cell line I use to confirm it has a myofibroblast phenotype. Typically myofibroblasts test positive for Vimentin and Smooth muscle actin but are negative for desmin. I am preferably not looking to purchase large amounts of either antibody, is there any way I can purchase smaller amounts than the advertised quantities?

Question

Dear Abcam,



Hi, I'm trying to detect fibrosis on a rat heart tissue that has undergone ischemia reperfusion injury.

I wanted to know if your anti-vimentin antibody ab137321 works on rat heart tissue for IHC-P and WB.

Some antibodies that say they cross react with rat, did not react with the heart so I wanted to make sure this antibody will work

Also, for anti-F4/80 ab74383, it says it works with WB but does it also work with IHC-P?

Do you have any references, articles that have stated use of these antibodies on rat heart tissue?

Answer

Thank you for your enquiry and your interest in our products.

I can confirm that both antibodies have been tested for cross-reaction with rat so they recognize this species. However, neither of them have been specifically used on rat hear tissue sections.

It would be worth checking some published papers using PubMed if F4/80 and Vimentin are widely used as good markers for investigating fibrosis on heart tissue. If these proteins are expressed at detectable levels, then ab74383 and ab137321 can be used in this study.
