ICC/IF Use a concentration of 0.25 - 2 µg/ml.
IHC-P 1/700. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Target

  • Function

    Oocyte-specific protein tyrosine kinase that phosphorylates and inhibits CDK1 and acts as a key regulator of meiosis during both prophase I and metaphase II. Required to maintain meiotic arrest in oocytes during the germinal vesicle (GV) stage, a long period of quiescence at dictyate prophase I, by phosphorylating CDK1 at 'Tyr-15', leading to inhibit CDK1 activity and prevent meiotic reentry. Also required for metaphase II exit during egg activation by phosphorylating CDK1 at 'Tyr-15', to ensure exit from meiosis in oocytes and promote pronuclear formation.

    Answer

    Thank your for your inquiry.

    The immunogen of ab121943 shows only 82% homology with mouse. We generally expect a cross reactivity from 85% homology. This is a border line case and I cannot recommend using this antibody in this case since WB is also not tested.

    If there is a negative result it will not be clear if the antibody does not recognize mouse or if it is simply unsuitable for WB.

    Maybe you want to consider running a WB with human samples of doing IHC-P or ICC/IF with the mouse samples?

    I am sorry I did not have a positive answer for your on this occasion and hope this information is nevertheless helpful.
