    why is the actual band size different from the predicted?

  • ICC/IF image of ab27794 stained MCF7 cells. The cells were 4% PFA fixed (10mins) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab27794, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Thank you for your enquiry. I am sorry to hear that you have been having difficulties with this antibody. I have read your technical questionaire and I have a few comments. Firstly it would be most informative if you could provide me with the size of the band that you have been detecting using this antibody. Furthermore it would be very useful if you could provide me with a representative blot of this band if possible so that I can pass this onto my colleagues in the lab. ab27794 was tested using rat kidney cells. On your technical questionaire you do not stipulate the type of rat cells that you have been using. I ask whether it is possible that you have been detecting a splice variant of WNT2 in the rat cells that you have been using? I would appreciate your comments on this. Given the band that you have been detecting, I would like to recommend that you rule out any potential cross reactivity of the secondary antibody by performing a no primary control experiment. I look forward to hearing from you.
