Target

  • Function

    Ligand for members of the frizzled family of seven transmembrane receptors. Probable developmental protein. Signaling by Wnt-7a allows sexually dimorphic development of the mullerian ducts.
  • Tissue specificity

    Expression is restricted to placenta, kidney, testis, uterus, fetal lung, and fetal and adult brain.
  • Involvement in disease

    Defects in WNT7A are the cause of limb/pelvis-hypoplasia/aplasia syndrome (LPHAS) [MIM:276820]; also known as absence of ulna and fibula with severe limb deficiency. LPHAS is a limb-malformation disorder characterized by various degrees of limb aplasia/hypoplasia and joint dysplasia.
    Defects in WNT7A are a cause of Fuhrmann syndrome (FUHRS) [MIM:228930]; also known as fibular aplasia or hypoplasia femoral bowing and poly- syn- and oligodactyly. Fuhrmann syndrome is a distinct limb-malformation disorder characterized also by various degrees of limb aplasia/hypoplasia and joint dysplasia.

      The expression profile observed is consistent with what has been described in the literature (PMID: 25978974, 30143647, 30770219). 

      Negative control: HeLa cells (PMID: 25978974).

      This blot was developed using a higher sensitivity ECL substrate.

      Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized HaCaT (human skin keratinocyte) and HeLa (human cervix adenocarcinoma epithelial cell) whole cells labelling Wnt7a with ab274321 at 1/50 dilution, followed by ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) antibody at 1/1000 dilution (Green). Confocal image showing strong cytoplasmic and weak nuclear staining in HaCaT cells. Low expression cell line: HeLa cells (PMID: 25978974). ab195889 Anti-alpha Tubulin mouse monoclonal antibody - Microtubule Marker (Alexa Fluor® 594) was used to counterstain tubulin at 1/200 dilution (Red). The Nuclear counterstain was DAPI (Blue).

      Secondary antibody only control: Secondary antibody is ab150077 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) at 1/1000  dilution.
