  • Function

    Plays a role in T-cell development and lymphocyte activation. Essential for TCR-mediated IL-2 production. Isoform 1 induces TCR-mediated signal transduction, isoform 2 does not.

    Phosphorylated on tyrosine residues upon T-cell antigen receptor (TCR) stimulation. Tyr-319 phosphorylation is essential for full activity.
  • Cellular localization

    Cytoplasm. Cell membrane. After antigen stimulation, isoform 1 concentrates at the immunological synapse and isoform 2 remains cytoplasmic. Co-localizes together with RHOH in the immunological synapse. RHOH is required for its proper localization to the cell membrane and cytoskeleton fractions in the thymocytes.

    10% SDS-PAGE gel.

  • Jurkat (human T cell leukemia cell line from peripheral blood) cells stained for ZAP70 (green) using ab227237 at 1/500 dilution in ICC/IF. Cells were fixed in 4% paraformaldehyde at RT for 15 minutes.

    Blue: Hoechst 33342 staining.

  • Paraffin-embedded U373 xenograft tissue stained for ZAP70 using ab227237 at 1/500 dilution in immunohistochemical analysis.

  • ZAP70 was immunoprecipitated from Jurkat (human T cell leukemia cell line from peripheral blood) whole cell extract with 5 µg ab227237. Western blot was performed from the immunoprecipitate using ab227237.

    Lane 1: Jurkat whole cell extract.

    Lane 1: Control IgG IP in Jurkat whole cell extract.

    Lane 2: ab227237 IP in Jurkat whole cell extract.
